Hiltonol, a dsRNA Mimic, Promotes NK Cell Anti‐Cancer Cytotoxicity Through TAZ Cytoplasmic Sequestration

چکیده

The evolutionarily conserved YAP/TAZ mechano-responsive transcription cofactors regulate development and tumorigenesis. Here, we show for the first time that human natural killer (NK) cells specifically express TAZ but not YAP, serves to limit NK cytotoxicity. synthetic double-stranded RNA (dsRNA), hiltonol, was able trigger cytoplasmic compartmentalization of TAZ, which increased Mechanistically, a low dose hiltonol enhanced actin-based intracellular contractility accompanied by an increase in active RhoA myosin light chain phosphorylation, conceivably through ERK1/2 activation-dependent ROS production. Importantly, showed dissociation LATS1 from actin upon activation associated with localization. Functionally, also reduced cell surface inhibitory receptors, e.g. KIR3DL1, c-Myc inhibition. Direct inhibition promoted cytotoxicity against K562 lymphoblast. We corroborated our findings coculturing hiltonol-pretreated breast lung cancer cells, demonstrated NK-mediated killing, occurred via cytoplasm facilitated Our pave way ex vivo rejuvenation immunotherapies, could involve cocktail inhibitor. TAZ promotes cell growth and inhibits Celastrol-induced cell apoptosis

Hippo pathway is a highly conservative signalling pathway related to the development of organisms, which has been demonstrated to be strongly linked to the tumorigenesis and tumour progression. As the major downstream effector of Hippo pathway, yes-associated protein (YAP), is a transcriptional activator of target genes that are involved in cell proliferation and survival. As an oncogene, YAP c...

The Hippo transducer TAZ promotes cell proliferation and tumor formation of glioblastoma cells through EGFR pathway

TAZ, a WW-domain-containing transcriptional co-activator, is important for development of various tissues in mammals. Recently, TAZ has been found to be overexpressed in some types of human cancers. However, the role of TAZ in glioblastoma remains unclear. In this study, we found that TAZ was overexpressed in prognostically poor glioblastoma patients. Through knocking down or overexpressing TAZ...

Rotavirus NSP4 protein as a viral biotoxin selectively promotes cytotoxicity

Abstract Background and Aims: The number of new cancer cases with considerable mortality is increasing worldwide. Since the inability of current therapies in treatment of patients and prevention the progress of tumors with fewer side effects, implementation of new methods is needed. Gene therapy has widespread systemic cytotoxic effects against tumor cells. Rotavirus NSP4 has been shown to...

Overexpression of FZD7 promotes glioma cell proliferation by upregulating TAZ

Gliomas are the most prevalent type of primary brain tumors in adults, accounting for more than 40% of neoplasm in the central nervous system. Frizzled-7 (FZD7) is a seven-pass trans-membrane Wnt receptor that plays a critical role in the development of various tumors. In this study, we detected high-level FZD7 expression in glioma and its overexpression was associated with advanced tumor stage...
